In Pennsylvania, the US routes were probably signposted starting in 1928.  At the time, three of these routes ended in Reading; each of them were marked on the map shown here.  During this timeframe (late 1920s and early 1930s), highways were commonly signed to a central location in the downtown area.  Although this map is not explicit about where these particular routes ended, highways in Reading would have likely been signed to Penn Square (the intersection of 5th Street and Penn Street):
